Western blot analysis of SirT1 expression in K562 whole cell lysates. The lane on the left is treated with the antigen-specific peptide.
Background:
Silent information regulator?1 (SIRT1), is a member of the sirtuin family of proteins, homologs of the Yeast Sir2 protein. Yeast SIRT1 is a nuclear protein that regulates epigenetic gene silencing and suppresses recombination of rDNA. Sirt-1 is defined as a nuclear protein involved in the molecular mechanisms of inflammation and neurodegeneration through the de-acetylation of many different substrates even if experimental data in mouse suggest both its cytoplasmatic presence and nucleo-cytoplasmic shuttling upon oxidative stress (1). Sirt-1 plays important functional roles in many biological processes causing various modifications of histone/protein acetylation status by several class I and II histone deacetylase (HDAC) inhibitors (2).
