Age-related decline of cardiac performance is often associated with oxidative stress and increased levels of free radicals, encompassing both reactive oxygen (ROS) and nitrogen species (RNS). Formation of RNS species is reported in acute myocardiac disorders such as heart failure. These species are metabolites and/or oxidation products of nitrogen monoxide (NO), such as nitrogen dioxide (NO2) and peroxynitrite (ONOO). A hallmark of these species is the conversion of tyrosine to 3-nitrotyrosine (3-NY).
Badrilla's monoclonal antibodies have been selected for their high affinity and specificity towards NY residues. They do not cross-react with tyrosine or tyrosine derivatives such as aminotyrosine, chlorotyrosine or phosphotyrosine, neither with nitroTrp which can also be generated in response to peroxynitrite. They also recognize NY residues in various sequences, as shown by their ability to recognise nitrated proteins including albumin, ovalbumin, insulin, hemoglobin, KLH and various cytoplasmic and mitochondrial proteins.
Tested applications: Western blot (1:2000 dilution). Reacts with Nitrotyrosine either free or incorporated in proteins such as human serum nitro albumin, nitro haemoglobin and nitro insulin.
